摘要

目的 了解合肥市一起急性胃肠炎疫情的病原分子特征。方法 采用实时荧光PCR(Real-time PCR)对13份肛拭标本同时进行札如病毒、诺如病毒、星状病毒核酸检测,检出的札如病毒核酸阳性标本采用传统RT-PCR扩增VP1基因片段,并测定基因序列,构建系统进化树。结果 Real-time PCR 检出8份札如病毒核酸PCR阳性标本,诺如病毒、星状病毒核酸结果均为阴性。1份RT-PCR扩增产物测序成功,基因序列比对和进化分析显示为札如病毒GⅡ.3基因亚型。结论 引起该起急性胃肠炎疫情的病原体为GⅡ.3型札如病毒。

Abstract

Objective To investigate the pathogenic molecular characteristics of an acute gastroenteritis epidemic in Hefei City. Methods The nucleic acids of sapovirus,norovirus and astrovirus were detected in 13 anal swab specimens by Real-time PCR.The detected sapovirus nucleic acid positive specimens were used traditional RT-PCR to amplify the VP1 gene fragment,and the gene sequence was determined to construct a phylogenetic tree. Results The results of real-time PCR showed that 8 samples of sapovirus nucleic acid were positive,while norovirus and astrovirus nucleic acid were negative.One RT-PCR amplification product was sequenced successfully.Sequence alignment and evolutionary analysis showed that it was a subtype of sapovirus GII.3. Conclusions The pathogen causing the outbreak of acute gastroenteritis was sapovirus GII.3 genotype.
