目的 根据MDCK细胞培养和鸡胚培养两种方法在分离乙型流感病毒中的对比研究,探讨提高流感病毒分离效率的方法。方法 收集淮南市流感监测哨点医院流感病毒核酸检测阳性的42份乙型流感病例的呼吸道标本,分别接种MDCK细胞和SPF鸡胚,通过红细胞凝集试验及血凝抑制试验,测定收获毒株(均为接毒一代的毒株)的血凝效价并分析毒株型别。结果 42份流感病毒核酸检测阳性乙型流感病例呼吸道标本,MDCK细胞法分离出毒株23份,鸡胚法分离毒株14份,分离率分别为54.76%和33.33%;相应毒株血凝试验血凝滴度达到1∶8及以上的分别占82.60%(19/23)和71.43%(10/14)。与鸡胚法相比,细胞法更为灵敏,细胞法培养血凝阴性鸡胚法均为阴性,两者是一致的。结论 MDCK细胞法和鸡胚法均可用于乙型流感病毒分离,两种方法有机结合应用,有利于提高流感病毒分离效率。
Abstract
Objective To explore the method of improving the isolation efficiency of influenza virus according to the comparative study between MDCK cell method and chicken embryo one in the isolation of influenza B virus. Methods A total of 42 respiratory specimens of influenza B positive cases detected by nucleic acid test from sentinel hospital of Huainan were inoculated into MDCK cells and chicken embryos respectively. The hemagglutination titer of the harvested strains (all of the first generation strains) and the type of the strains were determined by erythrocyte agglutination test and hemagglutination inhibition test. Results 23 strains of influenza B virus were isolated by MDCK cell method and 14 strains were isolated by chicken embryo method. The isolation rates were 54.76% and 33.33% respectively. 82.60% (19/23) and 71.43% (10/14) of the corresponding strains had hemagglutination titers of 1:8 and above,respectively.Compared with the chicken embryo method,the cell method was more sensitive,and the method of culturing chicken embryo with negative hemagglutination by cell method was negative,both of them were consistent. Conclusion Both MDCK cell method and chicken embryo one can be used for the isolation of influenza B virus. The combination of the two methods can improve the isolation efficiency of influenza virus.
