摘要
目的 应用PWAX固相萃取柱-高效液相色谱法检测8种合成着色剂,探讨该方法的可行性。方法 样液经PWAX SPE小柱净化,一次性提取净化检测食品中柠檬黄、苋菜红、酸性靛蓝、胭脂红、日落黄、诱惑红、亮蓝和赤藓红合成着色剂,以甲醇和乙酸铵为流动相,在C18色谱柱上分离,梯度洗脱,400~800 nm多波长PDA检测,外标法定量。结果 在0.2~20 g/ml浓度范围内,各目标物线性相关系数均>0.99988,最低检测浓度为0.020 μg/ml,方法最低检出限0.020 mg/kg,不同样品基质加标回收率为73.0%~102.7%,RSD为0.46%~5.35%。结论 该方法吸附解吸附效果均好、试剂用量少、操作简便,HPLC法对8种着色剂色谱分离度好,方法回收率和精密度满意,适合不同食品样品中多种合成着色剂检测分析。
Abstract
Objective To test eight kinds of synthetical colorants by PWAX solid phase extraction –HPLC, and discuss the feasibility of this mothod. Method The liquid samples were purified by PWAX SPE column, disposable extraction and purification the synthetical colorants (lemon yellow, amaranth, indicarmine, carmine, sunset yellow, allura red, brilliant blue, erythrosine) in foods,separated in the C18 column with methanol and ammonium acetate as mobile phase, detected by 400-800nm multi-wavelength PDA method after gradient elution.Quantified by external standard method. Results Within the concentration range of 0.2-20 μg/ml, the correlation coefficients of all the target substances were more than 0.99988, the lowest concentration of the test liquid was 0.020 μg/ml, the lowest detection limit was 0.020mg/kg.Recovery rate of different sample matrices were 73.0%-102.7%,RSD was 0.46%-5.35%. Conclusion The method had manig advantages such as well pigment adsorption and desorption effects,reducting the amount of reagents, simplicity and conveniency, good separation degree etal.the chromatographic separation of eight kinds of coloring agents was good by HPLC method.The recovery and precision of the method were satisfactory.It was suitable to detect and analyze various synthetic colorants in different food samples.
