摘要

目的 了解合肥市某工地一起急性胃肠炎聚集性疫情的病原及其基因型。方法 采用实时荧光PCR方法对16份肛拭子标本和7份水样标本进行诺如病毒、札如病毒和星状病毒核酸筛查,对诺如病毒核酸阳性标本采用RT-PCR扩增RdRp-VP1区基因,并进行基因序列测定,在线比对确定病毒基因亚型,运用MEGA软件构建基因进化树,进行基因比对和进化分析。结果 14份肛拭子和2份水样标本诺如病毒核酸阳性,札如病毒和星状病毒核酸均阴性,获得9条诺如病毒RdRp-VP1区基因序列,均为GⅡ.P17-GⅡ.17型。结论 这是一起因饮用水污染GⅡ.P17-GⅡ.17型诺如病毒引发的急性胃肠炎暴发疫情。

Abstract

Objective To understand the pathogen and genotype of an acute gastroenteritis cluster epidemic at a construction site in Hefei City. Methods Real time fluorescent PCR was used to screen the nucleic acid of norovirus,sapovirus and astrovirus in 16 anal swabs and 7 water samples.RT-PCR was used to amplify the gene of RdRp-VP1 region in norovirus nucleic acid positive samples.Gene sequencing was performed and viral genotype subtypes were determined by online comparison.MEGA software was used to build a gene evolution tree for gene comparison and evolution analysis. Results 14 anal swabs and 2 water samples were positive for norovirus nucleic acid,while both sapovirus and astrovirus nucleic acid were negative.9 gene sequences in the RdRp-VP1 region of norovirus were obtained,all of which were GⅡ.P17-GⅡ.17 type. Conclusion This is an acute gastroenteritis epidemic caused by drinking water contamination with norovirus type GⅡ.P17-GⅡ.17.
