摘要

目的 了解芜湖市首例人感染H7N9禽流感病例病原情况。方法 采集病例呼吸道标本并提取RNA,进行流感病毒特异性基因的PCR扩增,毒株提交CNIC进行基因测序。结果 患者呼吸道吸取物的样本FluA 和H7N9基因核酸扩增结果阳性。病毒HA和NA基因序列与其他已知H7N9禽流感分离株序列具有高度的一致性(HA:99.4%~99.8%;NA:99.2%~100.0%),HA基因核苷酸相似性最高毒株为KM054796-A/chicken/Jilin/2014,NA基因相似度最高的毒株为KP414837-A/chicken/Jiangxi/2014。HA和NA基因遗传进化关系最近的都为毒株KP414837-A/chicken/Jiangxi/2014。结论 芜湖市首例人感染H7N9禽流感病例的病毒株HA和NA序列均与已知H7N9禽流感病毒高度同源。

Abstract

Objective To Understand the pathogens of the first case of human H7N9 infections. Methods The respiratory specimens were collected from a case with pneumonia of unkno-wn cause reported in Wuhu city, Viral RNA was extracted from the specimens and detected on specif-ic genes by real-time RT-PCR, The specimens were submitted to CNIC for gene sequencing. Results FluA virus and specific H7N9 subtype from the resp,ratory tract estract were positive, The nucleotide acid sequences of HA and NA gene were highly similar with ot-her known avian isolate strain sequences(HA: 99.4%~99.8%;NA: 99.2%~100.0%), The highest similarit-y of nucleotide sequence of HAgene was KM054796-A/chicken/Jilin/2014,The highest similarity of NA gene was KP414837-A/chicken/Jiangxi/2014. there was the closest evolutionary relationships with KP414837-A/chicken/Jiangxi/2014. Conclusion The viral HA and NA sequences of the first case of human infection with H7N9 avian influenza in wuhu city were highly homologous to the known H7N9 avian influenza virus.
